目的检测长链非编码RNA AK125512在肝癌、癌旁肝组织中的表达情况,探讨其与临床病理指标的关系及作为预测肝癌患者术后预后标记物的潜在价值。方法从本课题组已完成表达谱芯片(GSE54238)中筛选明显差异表达lnc RNA-AK125512,采用实时荧光定量PCR(q RT-PCR)法检测lnc RNAAK125512在肝癌、癌旁肝组织中的表达;x2检验分析肝癌组织中lnc RNA-AK125512表达量与临床病理指标相关性;应用Log-rank检验和COX回归模型分析肝癌组织中lnc RNA-AK125512表达量与肝癌患者术后预后的关系。结果 lnc RNA-AK125512在肝癌组织中较癌旁肝组织明显低表达(P〈0.0001);肝癌组织中lnc RNA-AK125512的表达水平与Edmondson分级(P=0.022)、肿瘤包膜完整与否(P=0.005)、血AFP(P=0.018)明显相关;肝癌组织中lnc RNA-AK125512相对低表达患者肝癌切除术后无瘤生存期(P=0.0041)及总体生存期(P=0.0008)均显著缩短。肝癌组织中lnc RNA-AK125512相对低表达是影响肝癌患者术后无瘤生存期(P=0.037)、总体生存期(P=0.003)的一个独立危险因素。结论 lnc RNA-AK125512有望成为预测肝癌患者术后预后的一个新的潜在标记物。
Objective To investigate the expression of the long non-coding RNA AK125512 in hepatocellular carcinoma(Hcc) and its clinical significance. Methods From the lncRNA microarray(GSE54238) which we completed before, lncRNA-AK125512 was selected because of its obvious expression difference. The realtime fluorescence quantitative PCR(qRT-PCR) was performed to detect the expression difference of lncRNAAK125512 between HCC and its adjacent normal tissue. Then, the correlation between the expression of lncRNA-AK125512 in HCC and the clinicopathological characteristics was analyzed by Chi-square test. What's more, the potential of lncRNA-AK125512 in predicting the prognosis of patient with HCC after operation was assessed by Log-rank test and Cox's proportional hazards regression model. Results The expression of lncRNAAK125512 in HCC was markedly lower than that in its adjacent normal tissue(P〈0.0001). Moreover, the expression of lncRNA-AK125512 in HCC was significantly correlated with Edmondson classification(P=0.022),complete tumor capsules(P=0.005), alpha fetoprotein(AFP) serum levels(P=0.018). The patients with lower expression of lncRNA-AK125512 in HCC had a statistically shorter tumor-free survival time and overall survival time(P=0.0041, 0.0008).What's more, the lower expression of lncRNA-AK125512 in HCC was an independent risk factor for the tumor-free survival time(P=0.037) and overall survival time(P=0.003) of patients with HCC after curative hepatectomy. Conclusion lncRNA-AK125512 may be a new prognostic biomarker for hepatocel lular carcinoma after curative hepatectomy.
