目的 研究ezrin蛋白表达与青海汉族、回族胃癌易感性关系;同时研究ezrin对胃癌细胞形态、骨架和增殖的影响.方法 利用免疫组化对正常胃黏膜组织和胃癌组织进行ezrin蛋白表达检测,用Western blot蛋白印迹法检测两者ezrin蛋白表达水平,同时利用细胞转染及免疫荧光标记方法检测ezrin对胃癌细胞骨架调节的作用,而细胞增殖则用MTT方法进行检测分析.结果 ezrin蛋白在青海汉族、回族胃癌组织中表达比正常胃黏膜组织中高,具有显著性差异;同时ezrin蛋白表达水平与青海汉族、回族胃癌分化程度、淋巴结转移以及组织学分型有关,而和年龄、性别无关.此外,ezrin蛋白表达水平与青海汉族、回族民族无关,即无民族特异性.蛋白印迹法检测结果显示,肿瘤组织中ezrin蛋白表达水平比正常组织中较高.而细胞免疫荧光检测中,发现ezrin对细胞骨架、细胞形态及细微结构的调节起重要作用,同时MTT检测发现,ezrin转染的胃癌细胞增殖受到一定抑制.结论 ezrin蛋白在青海汉族和回族胃癌组织中表达水平比正常胃癌组织高,同时与胃癌分化程度、组织学分型和淋巴结转移有关;ezrin蛋白表达水平与汉族、回族不同民族无关.由于ezrin转染细胞在细胞形态、骨架和微绒毛等结构方面比对照组变化大,提示Ezrin蛋白与胃癌转染及侵移有关.
[Objective] To explore the relationship between the susceptibility and ezrin protein expression in gastric cancer of Qinghai Han- and Hui (muslim)-ethnic group. Meanwhile, we want to investigate the influences of ezrin on the cell morphology, cell polarization and cell proliferation of gastric cancer. [ Methods ] Immunohisto- chemicial staining is used to detect the expression of ezrin protein in normal gastric mucosa and gastric cancer tis- sues. Western blot is used to detect the ezrin expression level in both normal and gastric cancer tissues. Cell trans- fection and immunofluoreseent staining are used to observer the cell morphology change and cell polarization. Cell proliferation is analyzed by MT'F method. [ Results ] Ezrin protein expression is much higher in the gastric cancer than normal gastric mucosa in both Han- and Hui-ethnic patients of Qinghai Province, and the expression level ofezrin is assoeiated with differentiation grade, lymph node metastasis, and histological type of gastric eancer in both Hart- and Hui (muslim)- ethnics. Through Western blot analysis, it shows ezrin protein expression in normal gastric mucosa is lower than gastric cancer. Furthermore, the protein expression level of ezrin is not related with Han- or Hui (muslim)-ethnic, that means that ezrin expression in gastric cancer is not associated with ethnic speicificity. Ezrin induces cell morphological change, more polarization and microvilli development, and meanwhile it inhibits cell proliferation. [ Conclusions ] The expression of ezrin protein is higher in gastric cancer than normal gastric mu- cosa, and it is also associated with the differentiation grade, lymph node metastasis, and histological type of gastric cancer in both Han- and Hui (muslim)-ethnic groups of Qinghai Province. Ezrin protein expression is not related with the ethnic Han- or Hui (muslim)-ethnic in Qinghai Province. Compared with the control, ezrin stimulates cell morphological change, cytoskeleton remodeling and microvilli development
