目的观察防风对六味地黄丸干预实验性视网膜变性作用的影响,从药效学角度初步探讨防风的引经作用。方法将碘酸钠诱导的视网膜变性大鼠按随机数字表法随机分为3组:蒸馏水组,六味地黄丸组,六味地黄丸+防风组,自造模后第1天分别用蒸馏水、六味地黄丸药液、六味地黄丸+防风药液灌胃治疗。正常大鼠不予造模。常规喂养作为正常对照组。分别于治疗7天、14天时采用病理学及TUNEL方法观察各组视网膜病变程度。结果治疗第7天,六味地黄丸组和六味地黄丸+防风组的病变程度较蒸馏水组减轻。3组病变范围得分分别为0.54±0.5,0.29±0.6和0.92±0.5,六味地黄丸+防风组病变范围与蒸馏水组相比,差异有统计学意义(P〈0.05)。TUNEL检测六味地黄丸组、六味地黄丸+防风组和蒸馏水组RPE细胞的凋亡率分别为69.2%、40%和69.6%,其中六味地黄丸+防风组与蒸馏水组相比差异有统计学意义(P〈0.05)。治疗14天时,六味地黄丸组、六味地黄丸+防风组和蒸馏水组病变范围得分分别为1.25±0.4,1.21±0.3和1.46±0.3,3组相比差异无统计学意义(P〉0.05)。但TUNEL检测显示六味地黄丸组、六味地黄丸+防风组RPE凋亡细胞数较蒸馏水组减少,而且没有出现外核层凋亡,六味地黄丸+防风组RPE细胞的凋亡率与蒸馏水组相比差异有统计学意义(P〈0.05)。结论防风可以增强六味地黄丸对实验性视网膜变性的干预作用,提示防风在方中可能起到了引经药的作用。
OBJECTIVE To observe the contribution of Radix Saposhnikoviae to the intervention effect on experimental retinal degeneration treated by Liuwei Dihuang pills, and initially investigate the function of meridian-lead-in drug through pharmacodynamics perspective. METHODS Sodium iodate induced retinitis pigmentosa rats were divided randomly into three groups: distilled water group, Liuwei Dihuang pills group, Liuwei Dihuang pills + Radix Saposhnikoviae group, and gavaged with distilled water, Liuwei Dihuang pills, Liuwei Dihuang pills + Radix Saposhnikovicte respectively from day 1 after sodium iodate treatment. Ten normal rats were used as the normal control without any treatment.On day 7, 14 after treatment, histopathologic study and TUNEL test were performed to evaluate the retinopathy. RESULTS On day 7 after treatment, there was less retinopathy in Liuwei Dihuang pills group and Liuwei Dihuang pills + Radix Saposhnikoviae group than distilled water group. The lesion grades were 0.54±0.5,0.29±0.6 and 0.92±0.5, and the difference between Liuwei Dihuang pills + Radix Saposhnikoviae group and distilled water group was significant (P〈0.05). The apoptosis rates determined by TUNEL were 69.2%, 40%, 69.6% in the three groups, and the difference between Liuwei Dihuang pills + Radix Saposhnikoviae group and distilled water group was significant (P〈0.05). On day 14, the lesion grades were 1.25±0.4,1.21±0.3 and 1.46±0.3 respectively in Liuwei Dihuang pills group, Liuwei Dihuang pills + Radix Saposhnikoviae group and distilled water group, there was no significant difference among the groups. However, in TUNEL test, it was shown that there wasless apoptosis cells in the treament groups, and no apoptosis was found in the inner nuclear layer which was found in the distilled water group. The apoptosis rated in the Liuwei Dihuang pills + Radix Saposhnikoviaewas significantly less than that of distilled water group (P〈0.05).
