目的探讨环氧氯丙烷对组织工程心脏瓣膜构建中基质金属蛋白酶-1(matrix metalloproteinase-1 MMP-1)表达的影响。方法采用去垢剂和胰蛋白酶消化制备的脱细胞猪主动脉瓣膜支架作为对照组,用加用3%环氧氯丙烷处理24小时的去细胞支架材料作为实验组。将培养的人骨髓基质干细胞(human bone maiTow derived stroma cells BMSCs)种植于脱细胞支架上构建组织工程心脏瓣膜(tissue engineering heart valve 1EHV),分别行石蜡包埋切片、HE染色和扫描电镜观察TEHV的组织结构,并行逆转录-聚合酶链反应(RT—PCR)测定BMSCs分泌MMP-1的功能。结果BMSCs在实验组脱细胞瓣膜表面生长良好,MMP-1的表达比对照组降低。结论环氧氯丙烷处理的脱细胞猪主动脉瓣膜支架,可以抑制BMSCs的MMP-1表达。
Objective To evaluate the effects of epichlorohydrin on the expression of matrix metalloproteinase 1(MMP-1) in creating tissue engineering heart valves. Methods The porcine aortic valve leaflets were digested by using detergent and trypsin as controls, aeellularized by epiehlorohydrin for 24h as experimental groups, the cultured human bone marrow derived stroma cells (BMSCs) seeded onto the acellularized scaffolds, frequently analyzed by biochemical examination, scanning electron microscopy, histology, and detected the expression of MMP-1 by RT- PCR. Results BMSCs grew into the pores and formed a confluent layer in aeellularized scaffolds from experimental group, the expression of MMP -1 was lower than that controls. Conchusion The porcine aortic valve leaflets acellularized by epiehlorohydrin may inhibit the expression of MMP -1.
