目的 探讨变应性鼻炎(AR)患者特异性免疫球蛋白E(S-IgE)、白介素17(IL-17)、白介素2(IL-2)、白介素9 ( IL-9)和白三烯的表达水平及其与疾病活动度的相关性. 方法 AR患者90例,根据疾病活动度分为轻度组、中度组及重度组各30例,分别采用半定量PCR、免疫组化法检测外周血、鼻黏膜组织IL-17、IL-2、IL-9表达水平,采用ELISA检测S-IgE、白三烯表达水平,并分析其与疾病活动度的相关性. 结果 外周血、鼻黏膜组织中轻度组IL-2表达水平明显高于中、重度组(P〈0.05),重度组IL-9、IL-17表达水平明显高于轻、中度组(P〈0.05). 重度组外周血S-IgE、白三烯表达水平明显高于轻、中度组(P〈0.05). IL-2与AR严重程度呈负相关关系、IL-9、IL-17、S-IgE、白三烯与AR严重程度呈正相关关系(P〈0.05).结论 S-IgE、白三烯、IL-2、IL-9、IL-17在AR的发生发展中均发挥着一定的作用,其表达水平与AR的疾病活动度存在一定相关性,检测其表达水平对临床判断AR的炎症活动程度有一定价值.
Objective To investigate the expression levels of specific immunoglobulin E(S-IgE),interleukin-17(IL-17),interleukin-2 (IL-2),interleukin-9(IL-9) and leukotriene(LT) in patients with allergic rhinitis(AR),and to explore the correlation between the expressions of these cytokines and AR activity.Methods Ninety AR patients were divided into mild group,moderate group and severe group depending on the disease activity,30 cases for each group.The expression levels of IL-2,IL-9 and IL-17 in the peripheral blood and nasal mucosa tissues were detected using semi-quantitative PCR and immunohistochemistry respectively.And the expression levels of S-IgE and LT were determined by enzyme linked immunosorbent assay(ELISA).The correlation between the expressions of these cytokines and the activity of AR was analyzed. Results In the peripheral blood and nasal mucosa tissues,the expression level of IL-2 in the mild group was significantly higher than the level in the moderate or severe group(P〈0.05),and the expression levels of IL-9 and IL-17 in the severe group were significantly higher than the levels in the mild or moderate group(P〈0.05).The expression levels of S-IgE and LT in peripheral blood was significantly higher in the severe group compared to the levels in the mild or moderate group(P〈0.05).There was a negative correlation between the expression of IL-2 and the activity of AR,but a positive correlation was observed between the activity of AR and IL-9/IL-17/S-IgE/LT(P〈0.05).Conclusion S-IgE,LT, IL-2,IL-9 and IL-17 may be involved in the pathogenesis and progress of AR.And to some extent,the expression levels of these cytokines correlate with the disease activity of AR,and thus their detections are valuable to evaluate the inflammation activity of AR.
