硼酸换药对糖尿病小鼠创面愈合的促进作用及其对mRNA表达的影响

目的 探究4%硼酸溶液换药对糖尿病小鼠创面愈合的促进作用及其对创面mRNA表达的影响。方法 雄性8周龄C57BL/6J小鼠30只,随机分为糖尿病组与对照组,每组各15只,糖尿病小鼠模型用链脲佐菌素诱导制作。在小鼠背部制作3个同等大小的全层皮肤创面后,每日1次分别以生理盐水(A处理法)、4%硼酸溶液(B处理法)和0.5%碘伏+3%过氧化氢溶液(C处理法)对创面换药,记录创面愈合率,创面结痂和感染情况,在伤后3d、7d、14d断颈处死小鼠后取创面组织,进行组织总RNA提取和实时荧光定量PCR,检测创面转化生长因子β1(TGF-β1)、I型胶原蛋白mRNA的表达。结果 糖尿病组B处理法各时相点创面愈合率显著高于A、C处理法(均P〈0.05)。14d时糖尿病组硼酸处理法TGF-β1mRNA表达显著高于对照组的硼酸处理组(P〈0.01)。结论 硼酸的弱酸性能刺激创面TGF-β1活化,促进创面愈合。

英文摘要:

Objective To explore the effect of boric acid on wound healing and expression of mRNA in diabetes mice. Methods Thirty 8-week old male C57BL/6J mice were randomly divided into a diabetic group and a control group, each of 15. Diabetes was intro duced into the former group using streptozotocin and 3 full-thickness skin wounds of the same size were made on the back of all mice in both groups. Then 0.9 % normal saline, 4 % boric acid solution and 0.5% iodine+3% hydrogen peroxide were used respec tively to care the wound every day. The healing rate and infection rate were observed. The mice were decapitated and the wound tissues were harvested to conduct real time quantitative PCR and detect the mRNA expression of the transforming growth factor beta 1 (TGF-α1) and Collagen I 3, 7 and 14 days after the intervention. Results Among diabetic mice, those treated with the boric acid had significantly higher wound healing rate than those cared with the other two regimens at all time points (P〈0.05 fro all). Moreover, 14 days after the intervention, the mRNA expression of TGE-β1 of diabetic mice treated with boric acid was significantly higher than those controls treated in the same way. Conclusion Boric acid can stimulate the activation of TGF-β1 in wounds and pro mote wound healing.
