为研究舒泰麻醉对大鼠脑内源性H2S含量及其合成酶CBS的影响,借以探讨内源性硫化氢及其合成酶CBS与麻醉之间的关系。将24只Wistar大鼠随机分为对照组(C组),翻正反射消失组(Z1组)和翻正反射恢复组(Z2组)。各组于相应时间点取脑,使用亚甲蓝染色法和WesternBlot检测各脑区内源性H2S含量及CBS蛋白表达量。结果显示,Z1组脑干和海马内H2S含量较C组均显著降低,且脑干和海马内CBS蛋白表达量较C组显著降低,而丘脑内CBS蛋白表达量显著升高(P〈0.05);Z2组脑干内H2S含量较C组显著降低,而丘脑内CBS蛋白表达量较C组显著升高,且海马内H2S含量与CBS蛋白表达量较其他四个脑区均显著升高(P〈0.05)。综合实验结果表明,舒泰与脑中内源性H2S及CBS的生成具有一定相关性。
In order to study the effect of Zoletil on endogenous hydrogen sulfide (H2S)and cystathionine β-synthase (CBS)in rat different encephalic regions, and explore the correlation among them, 24 Wistar rats were divided randomly into control group (group C) , disappearance of righting reflex (group Z1 )and recovery of righting reflex (group Z2). The brain tissues were taken when reaching each time point.The endogenous H2S contents and CBS protein expression in different encephalic regions were ana- lyzed by methylene blue staining and western blot.The results showed that the H2S contents and CBS protein expression in group ZI were significantly decreased in brainstem and hippocampus, and the CBS protein expression was significantly increased in thalamus (P〈0.05). In group Z2, the H2S contents were significantly decreased in brainstem, and CBS protein expression was significandy increased. The H2S contents and CBS protein expression in hippocampus were significantly increase than those in other encephalic regions (P〈0.05).The results indicate that Zoletil is related with the emergence of endogenous hydrogen sulfide and cystathionine β-synthase in brain.
