摘要:目的观察盐酸奥芬米洛对实验性自身免疫性脑脊髓炎(EAE)模型大鼠的下丘脑一垂体一肾上腺轴(HPA轴)功能的影响。方法以1.0mg/kg盐酸奥芬米洛治疗EAELewis大鼠和肾上腺切除的EAEWistar大鼠,检测各组EAELewis大鼠和肾上腺切除的EAEWistar大鼠的皮质醇水平,同时观察肾上腺切除的EAEWistar大鼠的神经症状和Thl,Th2分泌型细胞因子水平。结果1.0mg/kg盐酸奥芬米洛可显著提高肾上腺切除的EAEWistar大鼠的血清皮质醇水平(P〈0.01),可显著降低Wistar大鼠血清γ-干扰素(IFN-γ)、肿瘤坏死因子-α(TNF-α)、白细胞介素-2(IL-2)、白细胞介素-4(IL-4)水平和IFN-γ/IL-4比值(P〈0.01)。结论盐酸奥芬米洛抑制EAE发生的作用途径可能是通过增强HPA轴的功能,提高大鼠皮质醇水平来抑制Thl分泌型细胞因子,从而调节Thl/Th2的失衡。
Objective To observe the effects of oxfenmino hydrochloric acid on the function of hypothalamus- pituitary-adrenal axis(HPAA) in rats with experimental autoimmune encephalomyelitis (EAE). Methods The EAE of Lewis rats and Wistar rats were orally with 1.0 mg/kg oxfenmino hydrochloric acid respectively. Detected the cortisol levels of rat EAE models, observed the neurological symptoms and tested secretion levels of Thl, Th2 cytokines in adrenalectomy Wistar rats of EAE. Results 1.0 mg/kg oxfenmino hydrochloric acid could significant- ly improved serum cortisol levels of adrenalectomy Wistar rats ( P〈0.01 ), and could also significantly decreased the levels of IFN-γ,TNF-α, IL-2, IL-4 cytokines and ration of IFN-γ/IL-4 in Wistar rats of EAE (P〈0.01). Conclu- sion Oxfenmino hydrochloric acid inhibiting outcome of EAE may be relative to activate the HPA axis and in- crease cortisol levels which inhibited the Thl cytokine secretion, then regulate the balance of the Thl/Th2 cyto- kines.
