长颚斗蟋和迷卡斗蟋的遗传差异对于蟋蟀遗传多样性,分类鉴定等有重要意义。通过PCR技术扩增测定长颚斗蟋和迷卡斗蟋的mt DNA 16S r RNA、COⅠ与Cyt b部分基因序列。数据分析表明:长颚斗蟋和迷卡斗蟋16S r RNA,COⅠ和Cyt b的种间序列相似度分别约为91.5%、87.0%和85.5%。长颚斗蟋种内16S r RNA、COⅠ和Cyt b基因序列相似度分别为99.8%、98.9%和98.8%;迷卡斗蟋种内16S r RNA、COⅠ和Cyt b基因序列相似度分别为98.4%、98.4%、97.2%。研究结果表明:长颚斗蟋与迷卡斗蟋的种间遗传差异明显大于种内遗传差异;16S r RNA、COⅠ、Cyt b基因序列适合作为长颚斗蟋和迷卡斗蟋遗传差异研究及分类鉴定的分子标记;COⅠ和Cyt b基因较16S r RNA基因有更快的进化速度,其种间遗传差异也更为明显。
Genetic differences of Velarifictorus aspersus and Velarifictorus micado is of great significance for the genetic diversity analysis, classification and identification of crickets. PCR amplification was used to de-termine partial gene sequences of mt DNA 16 S r RNA, CO Ⅰ and Cyt b of Velarifictorus aspersus and Velarifictorus micado. Data analysis showed that the interspecific sequence similarity of 16 S r RNA, CO Ⅰ and Cyt b of Velarifictorus aspersus and Velarifictorus micado was approximately 91.5%, 87.0% and 85.5%. Intraspecific genetic analysis revealed that the sequence similarity of 16 S r RNA, CO Ⅰ and Cyt b of Velarifictorus aspersus was respectively 99.8%, 98.9% and 98.8% and that of Velarifictorus micado was respectively98.4%, 98.4% and 97.2%. According to the results, The interspecific genetic differences between Velarifictorus aspersus and Velarifictorus micado are more significant than their intraspecific genetic differences. The gene sequences of 16 S r RNA, CO Ⅰ and Cyt b can be used as molecular markers for the genetic difference research, classification and identification of Velarifictorus aspersus and Velarifictorus micado; CO Ⅰ and Cyt b have faster evolution than 16 S r RNA as well as more significant interspecific genetic differences.
