目的探讨冠心病患者外周血中活化T细胞、辅助性T细胞亚群(Th1、Th2)及CXC趋化因子受体3(CxCR3)与冠脉斑块稳定性的关系。方法79例冠心病患者分为三组:稳定型心绞痛组(A组,24例)、不稳定心绞痛组(B组,20例)、急性心肌梗死组(C组,35例)。另选健康体检者15例为对照组(D组)。采用流式细胞术检测四组外周血中活化T细胞百分比,实时荧光定量PCR检测Thl、Th2及CXCR3的表达。结果B组、C组外周血活化T细胞百分比、Thl细胞mRNA和CXCR3mRNA表达水平明显高于A组、D组(P〈0.05)。A组、B组、C组Th2细胞mRNA表达水平低于D组(P〈0.05)。结论活化T细胞水平、Thl/Th2比例失衡及CXCR3高表达提示冠脉斑块不稳定。
Objective To investigate the relationship between the stability of coronary atherosclerotic plaque and the level of activated T cell percentage, Thl/Th2 subsets and the expression of CXC chemokine receptor 3 in the patients with coronary arterial disease(CHD). Methods Sewenty- nine CHD patients were divided into three groups of A(24 cases, with stable angina pectoris), B(20 cases,with unstable angina pectoris) and C(35 cases, with myocardial infarction). Fifteen healthy people(group D) were taken as the controls. The activated T cell percentage was detected with flow cytometry. Thl,Th2 and CXCR3 mRNA were measured with RT-PCR. Results The percentage of activated T cell, the expressions of Thl rnRNA and CXCR3 mRNA were significantly higher in groups of B and C than those in groups of A and D (P〈0. 05). The expression level of Th2 mRNA was significantly lower in groups of A,B and C than that in group D(P〈0. 05). Conclusion Activated T cell, imbalanced ratio of Thl to Th2 and high expression of CXCR3 indicate an unstable coronary atherosclerotic plaque.
