目的:探讨干扰素联合利巴韦林对慢性丙型肝炎患者外周血中CD8^+T细胞亚群功能的影响。方法:密度梯度离心法从慢性丙型肝炎患者外周血中获取外周血单个核细胞,体外给予干扰素和(或)利巴韦林处理。用流式细胞术检测外周血单个核细胞中CD8^+T细胞分泌干扰素γ和穿孔素(Perforin)的水平;用实时荧光定量聚合酶链反应检测外周血单个核细胞中干扰素相关基因IRF7和IFIT1的表达。结果:细胞内因子检测发现干扰素联合利巴韦林处理能增加CD8^+T细胞中效应性T细胞分泌干扰素γ和Perforin的水平(P〈0.05),同时能增加中心记忆性T细胞和效应记忆性T细胞分泌Perforin的水平(P〈0.05);基因转录水平的检测发现干扰素联合利巴韦林处理后干扰素调节基因IFIT1的表达增加(P〈0.05)。结论:慢性丙型肝炎患者的外周血单个核细胞体外经干扰素联合利巴韦林处理后,CD8^+T细胞的杀伤功能增强。
Aim: To evaluate the function of CD8+T cells from patients with chronic hepatitis C virus after treated with interferon plus ribavirin. Methods: Peripheral blood mononuclear cells were isolated from whole blood by density gradient centrifugation and seeded into 24-well plate before treatment with interferon and( or) ribavirin. The secretion of IFN-γ and Perforin was detected by flow cytometry. Real time PCR was used to detect the expression of interferon regulatory factors IRF7 and IFIT1. Results: Compared with control,being treated with interferon plus ribavirin could increase the levels of INF-γ and Perforin secreted by effective T cells( P〈0. 05). In addition,Perforin secreted by effect memory T cells and centre memory T cells also increased( P〈0. 05). The expression of interferon regulatory factor IFIT1 up-regulated simultaneously( P〈0. 05). Conclusion: Interferon plus ribavirin may enhance the function of CD8+T cells.
