目的:观察N-乙酰-D-乳糖胺联合抗CD28单克隆抗体对细胞因子诱导的杀伤细胞(CIK细胞)增殖及杀伤功能的影响。方法:提取32例肺癌患者的外周血单个核细胞,分为对照组、抗CD28单克隆抗体组、N-乙酰-D-乳糖胺组及抗CD28单克隆抗体和N-乙酰-D-乳糖胺联合组(联合组),依据CIK细胞培养流程培养和分组处理后于第4、8、10、12、14天测定各组细胞增殖,第14天测定IL-2、IL-10、γ干扰素及肿瘤坏死因子-α的分泌及对K562细胞的杀伤作用。结果:抗CD28单克隆抗体、N-乙酰-D-乳糖胺单独作用均能促进CIK细胞增殖及IL-2及肿瘤坏死因子-α的分泌,增加效应细胞(CD3^+CD8^+细胞)的比例,增强CIK细胞对K562细胞的杀伤能力(P〈0.05),但二者联合没有协同效应(P〉0.05)。结论:N-乙酰-D-乳糖胺与抗CD28单克隆抗体均能增强CIK细胞增殖及杀伤功能,但二者无协同作用。
Aim:To study the influence of LacNAc with or without anti-CD28 mAb on the proliferation and function of CIK cells from patients with lung cancer .Methods:The peripheral blood was collected from 32 patients with lung cancer . The monocytes were extracted and divided into four groups after suspension , control group,anti-CD28 mAb group,LacNAc group,and anti-CD28 mAb combined with LacNAc group(combined group),then cultured as CIK technological process and treated accordingly.The ability of proliferation was measured on the day of 4th, 8th, 10th, 12th, and 14th.IFN-γ,TNF-α, IL-2 and IL-10 secreted by CIK cells were measured on the 14th day.Phenotypic analysis via flow cytometry and anti-tumor affection on K562 cells via CCK-8 assay were performed on the 14th day.Results:The proliferation of CIK cells was signif-icantly enhanced by LacNAc or anti-CD28 mAb.The cytokine secretion,ratio of effector cells(CD3^+CD8 ^+cells) and kill-ing efficiency of CIK cells all improved after being treated by LacNAc or anti-CD28 mAb(P〈0.05),but there was no in-teraction between LacNAc and anti-CD28 mAb(P〉0.05).Conclusion:It is indicated that CIK cells treated with LacNAc or anti-CD28 mAb exhibit a potential in cytotoxic activity against tumor cells , but there is no interaction between them .
