目的建立金顶侧耳中烟酸含量的测定方法。方法采用RP-HPLC法,色谱柱为C18柱,流动相为甲醇-0.004mol·L-1庚烷磺酸钠-乙酸(5:95:0.2),检测波长为263nm。结果烟酸对照品0.08~0.40μg与峰面积呈良好的线性关系(r=0.9999,n=5),样品的平均回收率为103.84%,RSD=2.80%(n=9)。结论所建方法简便、准确、重复性好,可测定金顶侧耳中烟酸的含量。
OBJECTIVE To establish an HPLC method for the determination of nicotinic acid in Pleurotm citrinopileatus Sing..METHODS Ionpair reversed-phase(RP) high performance liquid chromatography(HPLC)was performed using a C18 column with methanol-0.004 mol·L-1 heptane mahogany acid sodium-acetic acid(5:95:0.2)as mobile phase.The detection wavelength was set at 263 nm.RESULTS The calibration curve of nicotinic acid was linear from 0.08 to 0.4 μg(r = 0.9999).The average recovery was 103.84% with RSD of 2.80%.CONCLUSION This method was proved to be accurate and simple.And it can be selected to control the quality of Pleurotm citrinopileatus Sing.
