结合区段灌注技术,以万古霉素为手性选择剂构建毛细管电泳手性分离体系,研究了氨基酸对映异构体的分离。选择的最佳分离条件为:①含3mmol·L-1万古霉素和2mmol·L-1 HDB的pH7为200mmol·L-1硼酸缓冲溶液的分离区段;②灌注长度为压力3.45kPa,时间2.5min。以含2mmol·L-1 HDB的pH7为200mmol·L-1硼酸缓冲溶液为运行缓冲溶液对19种氨基酸衍生物进行手性分离。试验结果显示其中15对氨基酸对映异构体得到了良好分离。
Separation of enantiomers of amino acids was studied by the capillary electrophoretic chiral separation system established on the base of combined use of segmented filling technique and vancomycin as ehiral selector. The optimum separation conditions were selected as follows: (1) a solution of 200 mmol·L-1 H3BO3 buffer of pH 7 containing 3 mmol·L-1 vancomycin and 2 mmol ·L-1 HDB was placed in the separation segmented section with the H3BO3 solution of HDB as conveying solution; (2) the length of segmented filling was kept under a pressure of 3. 45 kPa through a time of 2. 5 min. As shown by the experimental results, 15 pairs of the enantiometric amino acids among the 19 derivatives of amino acids tested were well separated.
