目的 比较吴茱萸碱(Evo)、吴茱萸次碱(Rut)和吴茱萸总碱(Eta)对血管紧张素Ⅱ(AngⅡ)诱导大鼠心肌细胞肥大的抑制作用。方法 将培养3 d后的新生SD大鼠原代心肌细胞随机均分为对照组、模型组、Evo组、Rut组和Eta组;采用Image-Pro Plus 6.0图像分析系统测量单个心肌细胞的表面积,BCA法测定细胞的蛋白含量,实时荧光定量PCR检测心肌细胞肥大的标志性基因心房利钠因子(ANF)的表达。结果 AngⅡ能明显增加心肌细胞的表面积、细胞蛋白的含量和ANF的表达;Evo、Rut和Eta能明显抑制AngⅡ诱导的心肌细胞表面积的增加、细胞蛋白质含量的增多和ANF mRNA的表达,且抑制作用呈量效关系;但相同浓度的抑制效应无明显差别。结论 Evo、Rut和Eta可明显抑制AngⅡ诱导的心肌细胞肥大,但抑制效应无明显差异。
OBJECTIVE To compare the inhibitory effect of evodiamine (Evo), rutaecarpine (Rut) and evodia total alkaloids (Eta) in rat eardiomyocyte hypertrophy induced by angiotensin II ( Ang II) in vitro. METHODS The cultured cardiomyocytes from SD neonatal rats were randomly divided into control group, model group, Evo group, Rut group and Eta group. The cardiomyocyte surface area was detected by Image - Pro Plus 6.0 image analysis system, and protein level was measured by BCA and the mRNA expressions of atrial natriuretic factor(ANF) was elevuated by RT- PCR,respectively. RESULTS Ang II significantly increased the surface area, protein content and the ANF mRNA expression of cardiomyocytes. Evo, Rut and Eta significantly attenuated the changes induced by Ang II in a concentration - dependent manner. At the same concentration, the anti - cardiomyocyte hypertrophy effect of Evo, Rut and Eta had no significant difference. CONCLUSION Evo, Rut and Eta can significantly inhibit Ang II - induced cardiomyocyte hypertrophy. But, they have no difference in the inhibitory effect.
