目的:超声提取远志总皂苷,并用分光光度计进行总皂苷含量的测定。方法:超声提取远志总皂苷,以远志皂苷元为对照品,香草醛-冰醋酸-高氯酸为显色剂,采用分光光度法在585nm处测定远志总皂甙含量。结果:对照品在28-63μg范围内线性良好(r^2=0.9984),平均回收率为100.45%,RSD=1.59%(n=5)。超声提取和热回流提取的远志总皂苷含量分别为1.63%,1.50%。结论:超声提取法用时短,提取率高,能达到比传统回流法更理想的效果。分光光度法测定含量简便、快速、准确、可靠。
Objective: To discuss the use of spectrophtotmetry in the quantitative determination of total saponins in Polygala. Methods: The total saponins in Polygala tenuifoli were extracted by ultrasonic wave. Senegenin was used as standard control and vanillin-perchloric acid was used as chromogenic reagent. The detection wave length was 585 nm. Results: Absorbance shows linear relationship to the total saponins concentration in the range of 28-63μg at 585nm. The regression equation of concentration was obtained: A =0.0079 C +0.0689 (μg) with a correlation coefficient of r^2=0.9984. The average recovery of standard control was 100.45%, and the relative standard deviation (RSD) of precision test was 1.59% (n=5). The concentration of total saponins in Polygala extracted by ultrasonic wave and circumfluence was 1.63% and 1.50% respectively. Conclusion: Ultrasonic wave extraction is more efficient than the circumfluence extraction. Spectrophtotmetry is simple, rapid, sensitive and accurate for the quantitative determination of total saponins in Polygala.
