目的优化大孔树脂对野生中华猕猴桃根总三萜(ACP)的纯化工艺,并探究其抗肿瘤活性。方法以ACP百分含量为指标,来确定AB-8型大孔树脂对总三萜的提取工艺;并用CCK-8法检测ACP对人肠癌SW480、LOVO细胞,乳腺癌MCF-7细胞,肝癌SMMC-7721细胞增殖的作用,流式细胞术分析其对SW480细胞周期的影响。结果最佳工艺条件:上样液浓度为2.8 mg/mL,pH为7,流速为1 BV/h,洗脱体积为4倍树脂柱床体积(4 BV),洗脱流速为2 BV/h;ACP对细胞增殖有抑制作用并呈浓度和时间依赖性,并诱导SW480细胞发生G_1和S期阻滞。结论采用AB-8大孔吸附树脂可以简单有效地对ACP进行纯化,且纯化后的ACP可抑制肿瘤细胞增殖,可能是通过诱导细胞周期发生G_1和S期阻滞。
Objective To purify total triterpenes from Actinidia Chinensis Planch root by macroporous resin and to explore its anti - tumor activity. Methods The percentage of total triterpenoids from Actinidia Chinensis Planch root was taken as indexes to determine the extraction technology by AB - 8 macroporous resin. The effect of total triterpenoids on the proliferation rates of human colon cancer SW480 cells and LOVO cells, human breast cancer MCF - 7 cells, and human hepatoma SMMC - 7721 cells was assessed by CCK - 8 assay. Cell cycle of SW480 ceils was demonstrated by flow cytometry (FCM). Results The optimal conditions were as follow, the density of sample solution of 2. 8 mg/mL, pH = 7, flow rate of 1 BV/h, the loading volume of 4 BV, and the flow rate of 2 BV/h. The total triterpenes inhibited the proliferations of four tmnor cells in dose - and time - dependent manners. In addition, they significantly induced cell cycle arrested at the G1 and S phase. Conclusion Using AB -8 maeroporous resin would be a simple and effective method to purify total triterpenes from Actinidia chinensis Planch root. After treated with them, the proliferation of cancer cells were inhibited by inducing cell cycle arrested at the G1 and S phase.
