目的探讨导入单纯疱疹病毒胸苷激酶(HSVtk)自杀基因的骨髓间充质干细胞(BMSC)对胶质瘤细胞的抗肿瘤作用。方法将导入HSVtk自杀基因的BMSC细胞分别与多种脑胶质瘤细胞株(人类和大鼠)共同培养后,采用MTT法观察其对胶质瘤细胞增殖的抑制作用。体外侵袭性实验观察胶质瘤细胞培养上清对BMSCtk的趋化作用。结果我们发现在更昔洛韦的存在下,人类BMSCtk细胞对胶质瘤细胞具有很显著的体外抗肿瘤活性。与新鲜培养基比较,从人类和大鼠胶质瘤细胞获得的条件培养基都可以增强人BMSC向胶质瘤细胞的迁移活性。结论导入HSVt2基因的BMSC在“旁观者效应”的介导下能够产生很强的抗胶质瘤效应。
Objective To investigate the anti-tumor effect for human and rat glioma cel!s mediated by the bystander effect using bone marrow mes- enchymal stem cells ( BMSC ) as a "vehicle". Methods We valuated the anti-tumor ef[ect media, ed by the bystander effect using BMSCtk on glio- ma cells with a co-culture system. Results In vitro co-culture experiment showed a potent growth inhibition effects on glioma cells in the ganciclovir- containing medium. Migratory activity of the human BMSC towards the tumor cells was enhanced by the conditioned medium derived from both hu- man and rat glioma cells compared to fresh medium. Conclusion The bystander effect of BMSCtk cells is not cell type-specific, suggesting that the strategy could be quite feasible for clinical use.
