目的观察更年春方(GNC)对去卵巢大鼠皮肤组织胶原纤维的作用并探讨可能发生的机制。方法10~12月龄SD雌性大鼠60只随机分为假手术组(S组)、生理盐水组(N组)、GNC高剂量组(H组)、GNC低剂量组(L组)、尼尔雌醇组(E组),每组12只。除S组做假性手术外,其余各组均行双侧卵巢切除术。术后2W开始给予灌药,N组灌服生理盐水,H组灌服GNC 2ml/d、L组灌服GNC 1mg/d、E组灌服尼尔雌醇1ml,1次/w。3个月后测定大鼠血清雌二醇(E2)的水平,取大鼠背部皮肤组织进行组织学及免疫组织化学观察,测定皮肤组织羟脯氨酸(HYP)水平,RealTime—PCR定量分析皮肤组织表皮细胞生长因子受体(EGFR)、ER亚型mRNA的表达。结果GNC及尼尔雌醇均能提高大鼠皮肤组织HYP含量,提高皮肤组织EGFR、ERα、ERβmRNA的表达,血清E2水平均未升高。结论更年春方能增加去卵巢大鼠皮肤组织胶原纤维的表达,此作用与GNC影响胶原蛋白合成途径有关。其机制可能与提高皮肤组织ERmRNA的表达有关。
Objective To study the possible effects of Gengnianchun formula (GNC) on skin collagen in ovariectomized (OVX) SD rats and to investigate its possible mechanism. Methods 60 ten to twelve months old SD rats were randomly divided into sham-operated (S) ,saline (N), high (H) and low (L) dosage of GNC and nilestriol(E) groups. Except S group, the rats in the other groups ovarieeto- mized bilaterally 2 weeks ago received corresponding drugs ,respectively. Serum E2 levels were determined 3 months later in five groups. Skin tissue of the back area were got for histological and immunohistologieal study and measurement of hydroxyproline (HYP). Results PCR was performed to analyze the expression of epidermal growth factor receptor( EGFR), ERα ,ERβ of skin tissue. Results The amount of HYP significantly increased after treated with GNC or nilestriol. The mRNA level of EGFR, ERa, ERβ of skin tissue all enhanced dramaticly after treated with GNC or nilestriol. The serum E2 level had no significant increase. Conclusions GNC could improve the amount of skin collagen fibers of OVX SD rats by enhancing collagen biosynthesis. The mechanism may associate with the increased expression of ER subtype of skin tissue.
