目的:研究白香丹胶囊对焦虑模型大鼠血清及脑区脑源性神经营养因子(BDNF)水平的影响及焦虑症发病相关中枢机制。方法:采用足底电击法(Foot Shock,FS)诱发大鼠焦虑样行为,以旷场实验和高架十字迷宫实验对其进行行为学检测、ELISA检测大鼠血清BDNF含量、Western blot检测大鼠脑区BDNF蛋白的表达。结果:与空白组比较,模型组大鼠旷场实验总路程增加(P〈0.01),高架十字迷宫实验进入开放臂次数百分数(OE%)和开放臂停留时间百分数(OT%)均降低(P〈0.01),ELISA实验所测大鼠血清BDNF水平降低(P〈0.01),Western blot检测大鼠各脑区(左海马、右海马、额叶、下丘脑)BDNF蛋白表达水平降低(P〈0.01);与模型组比较,白香丹组和地西泮组大鼠运动总路程均降低(P〈0.05),OE%和OT%均升高(P〈0.05),BDNF水平均升高(P〈0.05),各脑区BDNF蛋白表达水平均升高(P〈0.05或P〈0.01)。结论:中药白香丹胶囊对足底电击法诱导大鼠焦虑样行为具有明显的拮抗作用,BDNF是其作用靶点之一。
Objective: To study the effect of Baixiangdan Capsule on levels of brain- derived neurotrophic factor( BDNF) in serum and brain regions and central mechanisms of pathogenesis of anxiety syndrome. Methods: Using foot shock method( Foot Shock,FS) to induce anxiety behavior in rats,the ethology of rats were detected by applying open field test and elevated plus-maze test. ELISA detection was for BDNF levels in rats' serum and Western blot for detecting expression of BDNF protein in rats' brain regions. Results: Compared with the normal group,the total path of open field test of the model group rats increased( P〈0. 01) and the open arm entry percent( OE%) and the open arm time percent( OT%) of elevated plus- maze test decreased( P〈0. 01). BDNF levels in rats' serum decreased by ELISA( P〈0. 01) and by Western blot BDNF protein levels in various brain regions( left hippocampus and right hippocampus,frontal lobe,hypothalamus) of rats decreased( P〈0. 01). Compared with the model group,the total path of the Baixiangdan Capsule group and the Diazepam group' s rats decreased( P〈0. 01),OE% and OT% increased( P〈0. 05),the levels of BDNF increased( P〈0. 05),expression of BDNF protein in each brain regions increased( P〈0. 05 or P〈0. 01). Conclusion: Traditional Chinese medicine Baixiangdan Capsule has obvious antagonism on rats with anxiety behavior induced by foot shock method and BDNF is one of the targets.
