为了研究矿井水中产氢菌的性质,以义马矿矿井水为实验原料,并在常温下保存备用,采用斜面培养技术和细菌分离技术研究该矿井水中产氢菌的生理特征、分子生态学特征,然后对该产氢菌进行了不同稀释度的富集培养,研究了该产氢菌的性质及生长特点.结果表明,分离出来的产氢菌在显微镜下观察呈现棒状,球状,并发出荧光;通过稀释度法对细菌进行分离培养,得出了这种细菌的调整期,对数期,稳定期,衰亡期的时间段,由此判断出细菌的生长周期;富集培养前后细菌数目明显增加,表明了利用此培养基培养产氢菌并使其生长繁殖的可行性.产氢菌的发现,对于目前促进绿色清洁能源的开发具有重要的意义.
Aimed at studying the properties of hydrogen bacteria in mine water,the mine water from Yima was chosen as experimental material,and preserved it at room temperature.This paper adopted the slant cultivation technology and bacteria isolation technology to investigate the type of hydrogen bacteria in the mine water and its physiological and molecular ecological characteristics.If conducted the enrichment culture of various dilutions to further analyze the properties and growth characteristics of this kind of bacteria.The results are as follows:(1) the isolated bacteria appear to be rod-like and globular and emit fluorescence through the observations under a microscope; (2) the bacteria are isolated and cultured by means of dilution,then the corresponding time of its adjustment phase,exponential phase,stationary phase,decline phase are obtained respectively,from which its growth cycle can further be inferred; (3) the number of bacteria increase significantly before and after enrichment culture,indicating that the use of this medium for the hydrogen production growth and reproduction of bacteria is feasible.The finding of this type of hydrogen bacteria is of great significance for the promotion of green nature and the development of clean energy.
