使用硅胶柱层析的方法对一株新菌发酵菊糖产果寡糖进行纯化,并采用高效液相色谱进行分析。为提高该菌发酵产果寡糖的量,从发酵所需要的菊糖浓度、氮源种类、氮源浓度以及无机盐进行条件优化,确定菌株最优发酵培养基为:菊糖35g/L,酵母浸粉0.5g/L,K2HPO40.8g/L,NaCl 0.5g/L,KNO30.5g/L。通过条件优化,果寡糖产量提高了60.88%。
Silica gel column chromatography was used to purify the fructooligosaccharide produced from inulin by a newly screened strain and the content of fructooligosaccharide was determined by HPLC. Fermentation conditions of the strain were optimized to increase the yield of fructooligosaccharide,including concentration of inulin,type and concentration of nitrogen source and inorganic salt.The optimal culture medium was determined as follows:inulin 35g/L,yeast powder0.5g/L,K2HPO40.8g/L,NaCl 0.5g/L,KNO30.5g/L,and the yield of fructooligosaccharide was increased by 60.88%.
