目的:观察背部循经推拿对亚健康模型大鼠海马脑源性神经营养因子(BDNF)基因和蛋白表达的影响,初步探讨背部循经推拿调理亚健康状态的作用机制。方法:60只Wistar大鼠随机分为正常组、模型组和推拿干预组,后两组予束缚应激,推拿干预组予背部循经推拿。实验结束后,采用实时荧光定量PCR方法检测大鼠海马BDNF mRNA表达,免疫印迹技术检测大鼠海马BDNF蛋白表达水平。结果:与空白组比较,模型组大鼠海马BDNF mRNA及其蛋白表达水平下降,差异有显著性(P〈0.05)。与模型组比较,推拿干预组大鼠海马BDNF mRNA及其蛋白表达水平均明显升高,差异有显著性(P〈0.05)。结论:背部循经推拿手法能上调亚健康模型大鼠海马区BDNF蛋白表达水平,这是推拿调治亚健康状态的作用途径之一。
Objective :To observe the influence of Chinese massaging along the meridians on the back on the expression of brain - derived neurotrophic factor (BDNF) gene and protein in the hippocampus of Sub - health Sample Rats. Methods : Sixty wister rats were divided into normal feeding group, model group and Chinese massag intervention group. The latter two groups were treated with restraint stress, beisdes, the massage group with massage. After the experiment, the expression levels of BDNF mRNA and protein of the hippocampus tissues were detected by Real time - PCR and Western blot methods, separately. Results : Compared with the control group, the expression levels of BDNF mRNA and protein in the hippocampus were significantly lower in the model group ( P 〈 0. 05 ). Whereas compared with the model group, the levels of BDNF mRNA and protein expression in the hippocampus were significantly increased in the Chinese massage intervention group( P 〈 0.05 ). Conclusion:Chinese massaging along the meridians on the back can effectively reverse restraint stress - induced down - regulation of BDNF mRNA and protein expression in the hippocampus of Sub - health Sample Rats, which may contribute to its mechanism in massage modulation effect to sub - health state by protecting the regeneration of neurons.
