目的通过改进大鼠心肌缺血/再灌注模型制备方法,提高模型成功率。方法雄性Wistar大鼠16只随机分为伪手术组(Sham组,n=8)和心肌缺血再灌注组(MI/R组,n=8)。缺血过程在人工机械通气条件下完成,再灌注过程在自主呼吸条件下进行,所有操作尽量不破坏动物组织结构,Evan’s蓝和TTC染色分析心肌梗死面积。结果MI/R组结扎心肌冠状动脉左前降支后心电图ST段弓背向上抬高0.4mV以上,Sham组手术前后无变化;MI/R组大鼠心肌梗死面积明显高于Sham组[(53.6±2.3)%vs(1.7±0.6)%,P〈0.001]。结论本方法制备大鼠心肌缺血/再灌注模型简便易行,心脏暴露好、结扎可靠,结果稳定。
Objective To improve the method of establishing in vivo myocardial ischemic reperfusion rat model and increase the success rate, Methods Sixteen male Wistar rats were randomly divided into sham group ( n = 8) and myocardial ischemie reperfusion group (MI/R, n - 8). In MI/R group, coronary occlusion for 30 min was followed by 5-hour reperfusion of left coronary artery. In the period of MI, rats breathed under the control of artificial respirator, while in the period of reperfusion the chest was closed and the natural respiration recovered. Results The infarct size in MI/R group (infarct area/area at risk) was (53.6 ± 2.3)% which was significantly higher than that in sham group. Conclusion The method is simple, convenient, and the experiment results are reliable.
