为了实现对细胞多个生理参数的同时记录,研制了一台自动分析仪,该仪器采用包含光寻址电位传感器(LAPS)、细胞电阻抗传感器(ECIS)、微电极阵列传感器(MEAs)3种细胞传感器的集成芯片及其检测系统.仪器集成了细胞生长环境控制系统和自动流动分析系统,能够满足多种不同的细胞生理实验需求.相比于以往的细胞生理检测仪器,具有多参数同时检测、多功能、自动灵活的特点.进行了细胞胞外酸化率变化和阿霉素作用下细胞阻抗和电位变化的实验,结果表明:分析仪能够对胞外离子、细胞阻抗、胞外电位进行同步、长时检测,可应用于药物评价、药物筛选等生物医学研究领域.
An automatic analysis instrument was developed to simultaneously record the multi-physiological parameters of the cells. The instrument employs an integrated chip which consists of light addressable po- tentiometric sensor (LAPS), electrical cell-substrate impedance sensor (ECIS) and microelectrode array sensor (MEAS), three different types of cell-based biosensors, and their detection systems. A cell growth environment control system and an automatic flow analysis system were integrated in the instrument, so it can meet different needs of cell physiology experiments. Compared with previous cell physiology testing in- struments, the new instrument comes to be multi-parameter, multi-function, automatic and flexible. An investigation of extracellular acidification rate change and doxorubicin inducing change of cell impedance and extracellular potential was carried out. Result shows that the instrument is capable of monitoring ex- tracellular ions, cell impedance and extracellular potential in synchronous and long-time. Thus it can be used in drug evaluation, drug screening and other areas of biomedical research.
