以金钗石斛(Dendrobium nobile)、流苏石斛(D.fimbriatum)和报春石斛(D.primulinum)为研究对象,研究了其在激素浓度配比下种子萌发情况,并对流苏石斛幼苗进行离体保存。结果表明:25℃时,Knudson C(KC)+BA2mg/L+N从0.2mg/L最利于金钗和流苏石斛原球茎诱导和幼苗分化;KC+NAA0.5mg/L时最适报春石斛原球茎诱导和幼苗分化。幼苗保存在15℃,每天12h光照(20001x),添加3%甘露醇(mannitol)的KC培养基中,10个月后存活率为96%。
Effect of different hormones combinations on the seed non-symbiotic germination of D. nobile, D. fimbriatum and D. primulinum also the in vitro conservation of D. fimbriatum seedings were studied in this paper. The results showed that KC+ BA2mg/L +NAA0.2mg/Lat 25℃ was the best for D. nobile and D.fimbriatum protcorm growth and organogenesis; KC+ NAA0.5mg/L at 25 ℃ was suitable for D. primulinum. Addition of mannitol ( 3.0%w/v ) to the medium and incubation at low temperature ( 15 ℃ ) resulted in higher survival rate ( 96% ) after 10 months of continuous storage.
