建立一种同时测定杨树花和杨树叶中豆甾醇、麦角甾醇、β-谷甾醇含量的反相高效液相色谱法。方法:以甲醇为流动相,测定杨树花和杨树叶中豆甾醇、麦角甾醇、β-谷甾醇的含量。结果:杨树花和杨树叶中3个植物甾醇的线性范围为0.00008-0.08μmol·m L-1,r〉0.9990。杨树花中麦角甾醇、豆甾醇、β-谷甾醇的加样回收率分别在92.6%-100.0%、93.9%-100.1%、94.0%-100.0%之间,RSD(n=6)分别为2.8%、2.2%、2.8%;杨树叶中各甾醇加样回收率分别在91.3%-100.0%、91.2%-100.0%、92.5%-100.1%之间,RSD(n=6)分别为2.7%、2.7%、2.5%。结论:该方法稳定、快速,精密度高,线性范围宽,适合杨树花和杨树叶中的豆甾醇、麦角甾醇、β-谷甾醇含量测定。
Objective: To establish a method for determination of the contents of three phytosterols(stigmasterol, ergosterol,β- sitosterol)from Flos Populi Canadensis and leaves of Populus canadensis Moench by RP- HPLC. Methods: With methanol as the mobile phase,HPLC method was adopted to determine contents of phytosterols in Flos Populi Canadensis and leaves of Populus canadensis Moench. Results: The three phytosterols had good linearity in their ranges of 0. 00008 - 0. 08 μmol · mL- 1 ( the coefficients of correlation were all over 0. 9990). The average recoveries of stigmasterol ,ergosterol β- sitosterol in Flos Populi Canadensis were 92.6% - 100.0% ,93.9% - 100. 1% ,94.0% - 100. 0% ,and RSDs(n =6)were 2. 8% ,2.2% ,2. 8% ,respectively;the av- erage recoveries( n = 6)of the three phytosterols in leaves of Populus canadensis Moench were 91.3% - 100.0% , 91.2% - 100. 0% ,92.5% - 100. 1% ,and RSDs were 2.7% ,2.7% ,2.5% ,respectively. Conclusion : The method is sensitive, accurate and reproducible, and reliable for the determination of phytosterols in Flos Populi Canadensis and leaves of Populus canadensis Moench.
