电针水沟穴对脑缺血再灌注大鼠自噬相关蛋白p62表达的影响

目的:探讨电针水沟穴对大鼠脑缺血再灌注(MCAO/R)损伤的保护作用及与自噬相关蛋白p62的关系。方法:线栓法复制大鼠局造性脑缺血再灌注模型。采用Bederson神经功能缺损体征评分法对MCAO/R大鼠进行神经功能评分;Western blot检测MCAO/R大鼠自噬相关蛋白p62的表达。结果:与假手术组比较,模型组大鼠有明显神经功能缺损体征,自噬相关蛋白p62的表达明显下调;与模型组相比针刺治疗组大鼠神经功能缺损体征明显减轻,自噬相关蛋白p62的表达明显上调。结论:电针水沟穴可能是通过拮抗自噬相关蛋白p62表达的下调减轻脑缺血再灌注损伤。

英文摘要:

Objective:To study the protective effect of electro- acupuncture at Shuigou(GV26) on cerebral ischemia referfusion injure in rats and discuss the relationship with autophagy -reiated Protein p62. Methods:Focal cerebral ischemia referfusion rats were induced by middle cerebral artery occlusion with thread embolus. By using of Bederson score method, the rats' neurological deficit scores were observed after MCAO/R injure. The level of autophagy - related Protein p62 was investigated by Western Blot. Results : The results suggest that the model group had obvious signs of neurological deficit and the expressions of autophagy - related Protein p62 were significantly decreased in comparison to that of the sham group. The results also showed that the neuro- logical deficit scores in treatment group were markedly decreased and the expressions of Autophagy - related Protein p62 were sig- nificantly increased in comparison to that of the model group. Conclusion:These results suggest that electroacupuncture at Shuigou (GV26) protects brain against ischemia reperfusion injury in rats through inhibiting the low -regulation of autophagy -related protein p62.
