沸水法提取得到的大粒车前子粗多糖经Sevag法脱蛋白后,再经葡聚糖凝胶层析柱反复纯化得到3个均一组分,即PLP-1、PLP-2、PLP-3,并对3个组分进行分子量及理化性质的测定.结果表明:PLP-1的相对分子量为3292494u,PLP-2的相对分子量为l725889u,PLP-3的相对分子量为1286790u;糖含量分别为82.08%、87.32%和79.18%;蛋白含量分别为0.54%、1.16%和0.72%;糖醛酸含量分别为17.46%、14.66%和20.13%.3个组分进行紫外光谱和红外光谱扫描结果显示,3个组分中均有蛋白质存在.
In this study, polysaccharides were isolated with boiling water from Plantago asiatica L., and then the protein in the isolates was removed by Sevag method. Successively, the polysaccharides were further purified using SephacrylTMS-400HR column gel permeation. The main components, named PLP (Plontago asiatica L. polysaecharides) -1, PLP-2, PLP-3 was obtained. The PLCP-1 was purified by SephacrylTM S-500 HR gel column. A homogeneous composition was obtained and defined as PLP-1; and the PLCP-3 was purified by SephacrylTM S-400 HR gel column. The homogeneous composition PLP-3 was obtained. The molecular weight of PLP-1, PLP-2 and PLP-3 were 3 292 494 u, 1 725 889 u and 1 286 790 u; The physicochemical properties of PLP-1, PLP-2 and PLP-3 were showed that content of sugar were 82.08%, 87.32% and 79.18%; the protein contents were 0.54%, 1.16% and 0.72%; while the content of uronic acid were17.46%, 14.66% and 20.13%; then the three components were identified by UV and IR spectrum.
