采用生物遗传监测系统,研究了涕灭威及其两个有毒代谢产物(涕灭威亚砜,涕灭威砜)对蚕豆及大蒜根尖细胞微核率的影响,以及对小牛胸腺DNA的加合作用.结果表明,两种植物根尖细胞微核试验测得的高浓度剂量诱导的微核率与对照组相比有显著性差异(P〈0.05),说明涕灭威、涕灭威亚砜及涕灭威砜可能对蚕豆及大蒜根尖细胞有致突变性.并且配对资料差异的显著性检验表明,蚕豆根尖微核试验的敏感性明显高于大蒜根尖微核试验的敏感性.涕灭威及其两个有毒代谢产物与小牛胸腺DNA结合引起吸收光谱波峰的移位,而且存在着剂量关系,说明涕灭威、涕灭威亚砜及涕灭威砜可能对小牛胸腺DNA造成加合作用。
Effects of Aldicarb and it's toxic metabolites, on the rates of micronucleus formation in root tip cells of Vicia faba and Allium sativum, and on the adduct actions to calf thymus DNA, respectively were studied in the present investigation. In some concentrations, micronucleus formation in root tip cells of Vicia faba and Allium sativum were induced by aldicarb, aldicarb sulfone and aldicarb sulfoxide. And with the increase of concentrations of aldicarb, aldicarb sulfone and aldicarb sulfoxide, the rates of micronucleus formation rosed in root tip cells. Compared with negative control group, results showed that in high concentration, rates of micronucleus formation in root tip ceils of Vicia faba and AUium sativum were remarkably different (p〈0.05), and the effects of aldicarb sulfoxide was more obvious than the effects of the others. The results indicated that aldicarb, aldicarb sulfone and aldicarb sulfoxide may exhibited carcinogenicity in root tip cells of Vicia faba and Allium sativum. And the distinct test of the partnership data difference showed that the sensitivity of micronucleus experiment in root tip cells of Vicia faba was obviously higher than that of AUium sativum. Using UV spectrum, the effects of Aldicarb and it's toxic metabolites on DNA were studied, the positions and intensions of absorbing peaks were changed, which implied aldicarb, aldicarb sulfone and aldicarb sulfoxide could bind to ctDNA to produce DNA adduct. Aldicarb and it's metabolites might result in the gene mutation and bring trauma to the DNA of the organisms through producing DNA adducts.
