目的探讨Wistar大鼠海马星形胶质细胞N-甲基-D-天冬氨酸受体(NMDAR)亚单位在β-淀粉样蛋白(Ap)25-35毒性作用下的表达变化特点。方法大鼠海马原代培养细胞,加Aβ25-35(10μmol/L)分别作用1h、24h后,应用免疫荧光方法检测对照组和加药组中NRl、NR2A和NR2B的表达情况(n=10)。结果对照组海马星形胶质细胞表达NRl、NR2A和NR2B,阳性点状颗粒主要分布在细胞胞体和突起上,在胞体部位分布密集,在细胞突起则散在分布。经Aβ25-35作用后,三者表达均显著增强(P〈0.05)。AIMh组和24h组相比,NR1无显著变化,NR2A、NR2B的表达随作用时间增加显著增强(P〈0.05)。结论正常状态下海马星形胶质细胞可表达NMDAR亚单位(NR1、NR2A和NR2B);Aβ25-35作用会引起NMDAR各亚单位表达显著增强,NR1的表达变化与NR2A和NR2B的变化表现出不一致性。
Objective To investigate the effects of β-amyloid protein ( Aβ25-35 ) on astrocytic N-methyl-D-aspartate receptor (NMDAR) subunits in the rat hippocampus. Methods Aβ25-35 (10μmol/L) was added into the primary cultured hippocampal cells for 1 hour and 24 hours. Immunofluorescenee technique was applied to investigate the changes of the expression of NR1, NR2A and NR2B between the control group and Aβ groups (n = 10). Results NR1, NR2A and NR2B were expressed in the hippocampal astrocytes. The positive puncta were mainly distributed in the soma and process. The expression was intensive in the soma and scattered in the process. Their expression was increased following Aβ25-35 treatment (P 〈 0. 05 vs control group). The expression of NR2A and NR2B were significantly increased (P 〈0. 05 ) between Aβ lhour group and Aβ 24hours group, while there was no significant changes in NR1 group. Conclusion The rat hippocampal astrocytes express NMDAR subunits ( NR1, NR2A and NR2B), and their expressions are significantly increased after AI5 treatment, while the variation tendencv of NR1 is different from the chanaes of NR2A and NR2B.
