目的观察赖氨酰氧化酶(LOX)在乳腺癌组织中的表达变化,探讨其与基质金属蛋白酶2(MMP-2)、基质金属蛋白酶9(MMP-9)、低氧诱导因子-1α(HIF-1α)的关系。方法分别应用RT—PCR和免疫组化法检测120例乳腺癌组织、癌旁乳腺组织及30例乳腺良性病变组织中的LOX mRNA和蛋白,分析乳腺癌组织中LOX表达与淋巴结转移的关系;用免疫组化法检测上述组织中的MMP-2、MMP-9、HIF-α蛋白,分析其在乳腺癌组织中表达与LOX的相关性。结果乳腺癌组织中LOX mRNA相对表达水平为0.44±0.08、LOX蛋白表达率为67.5%(81/120),均高于相应癌旁组织及乳腺良性病变组织(P均〈0.05);伴腋窝淋巴结转移的乳腺癌组织中,LOX表达高于无腋窝淋巴结转移者(P=0.000);MMP-2、MMP-9、HIF-1仅蛋白在乳腺癌中的表达阳性率为70.0%(84/120)、68.3%(82/120)、61.7%(74/120),均高于相应癌旁组织及乳腺良性病变组织(P均〈0.05);相关分析显示,LOX蛋白表达与MMP-2、MMP-9和HIF-1α蛋白之间呈显著正相关(r分别为0,450、0.423、0.462,P均〈0.05)。结论LOX在乳腺癌中高表达,并与淋巴结转移密切相关;它可能与MMP-2、MMP-9和HIF-1α相互作用,共同促进乳腺癌的侵袭、转移。
Objective To investigate the expression of lysyl oxidase (LOX) in breast caner tissue and its correlation with MMP-2, MMP-9 and HIF-1α. Methods The expression of LOX mRNA and protein were detected in 120 cases of human breast cancer tissue, the adjacent breast tissues, and 30 cases of breast benign lesion tissue by RT-PCR and immu- nohistochemical staining respectively. The relationship between the expression of LOX in the breast cancer tissue and lymph node metastasis was analyzed. MMP-2, MMP-9 and HIF-1α protein were detected by immnnohistochemical staining and the correlation of their expression with LOX in breast cancer was conducted. Results The expression level of LOX mRNA in breast cancer tissue was 0.44 ±0.08, and the positive rate of LOX protein was 67.5% (81/120) in breast cancer tissue, which were significantly higher than those of the adjacent breast tissues and breast benign lesion tissue (all P 〈 0.05 ). Expression of LOX was related to lymph node metastasis, and the expression of LOX in breast cancer tissue with lymph node metastasis was higher than that without metastasis ( P = 0.000). The positive rates of MMP-2, MMP-9 and HIF-1α protein were 70.0% (84/120), 68.3% (82/120) and 61.7% (74/120) in breast cancer tissue, which were significantly higher than those in the adjacent breast tissues and breast benign lesion tissue( all P 〈0.05). Correlation analysis showed that the expression of LOX was positively correlated with MMP-2, MMP-9 and HIF-1α ( r = 0. 450, 0.423, 0. 462, respectively ; all P 〈 0.05 ). Conclusion LOX is highly expressed and related with lymph node metastasis in breast cancer, and it may promote the invasion and metastasis of breast cancer by interacting with MMP-2, MMP-9 and HIF-1 α.
