目的观察抑制赖氨酰氧化酶(LOX)基因表达后对MDA-MB-231细胞的生物学特性的影响。方法将细胞分为实验组、空白对照组和阴性对照组,采用病毒转染法,应用噻唑蓝(MTT)法、细胞集落形成实验、流式细胞仪和细胞侵袭迁移实验检测结果。结果24、48、72、96h3 组细胞抑制率分别是(12. 73 ±3. 30)% .(33.93 ± 1. 99)% ,(52. 63 ± 1.04)% ,(63. 80 ±0. 70)%;0、0、0、0;(7. 60 ±6. 27)%、( 10. 30 ± 1. 49)%、(9.10 ±2. 69)%、(6. 03 ± 1. 21)%,差异有统计学意义(P 〈0.05)。细胞周期时相差异无统计学意义(P〉0.05)。细胞侵袭实验结果分别为47±2、100±1、88±2,差异有统计学意义(P〈0.05);细胞迁移实验结果分别为63 ±2,118 ±2,96 ±2,差异有统计学意义(P〈0. 05) 0结论干扰LOX基因的表达可抑制MDA-MB-231细胞的生物学特性。
Objective To observe the influence of RNAi against lysyl oxidase (LOX) gene on bi-ological features of MDA-MB-231 cells. Methods The MDA-MB-231 cells were transfected with lentiviralinterference vector ( LOX-RNAi-LV),and divided into three groups : the experimental group, the blankgroup and the negative group. The growth and proliferation of cells were determined by using methyl thiazoltetrazolium ( MTT) assay, cell colony formation and flow cytometry. The migration and invasion were ob-served by using transwell assays. Results The cell growth rate in the experimental group, the blank groupand the negative group at 24, 48, 72 and 96 h was (12. 73 ± 3. 30) % , ( 33. 93 ± 1. 99) %,(52. 63 ±1.04)% , (63. 80 ±0.70)%; 0,0,0,0; (7. 60 ±6. 27)%,(10. 30 ±1.49)%, (9. 10 ±2. 69)%,(6. 03 ± 1. 21 ) % , respectively ( P 〈 0. 05 ) . Cells invading between three groups was 47 ± 2; 100 ± 1;88 ±2 (p 〈0. 05) . Cell migration was 63 ±2; 118 ±2; 96 ±2 (p 〈0. 05). The difference of the cell cy-cle was not statistically significant (P 〉 0. 05). Conclusion After the LOX gene expression restrained,thecreature feature of MDA-MB-231 were effectively suppressed.
