利用甲基磺酸乙酯(EMS)诱变获得的拟南芥突变体库筛选过氧化氢(H2O2)敏感突变体,以期为研究氧化胁迫的分子机制提供遗传材料。从该突变体库中筛选出1株H2O2敏感突变体hpsl2(hydrogen peroxide sensitive12)。表型分析发现,hpsl2突变体植株矮小,果荚较短,并且在4mmol/LH202处理下hpsl2的子叶变绿情况明显低于野生型(wT),突变体的离体叶片在10mmol/LH2O2处理下相比wT表现出更为明显的衰老症状。进一步遗传学分析表明,该突变体为单基因隐性突变。
In order to investigate the mechanism of ROS signaling transduction in after-germination process,we screened hydrogen peroxide sensitive mutants from an EMS mutant library in Arabidopsis, to provide genetic material for study of molecular mechanism of oxidative stress. As a result,hydrogen pero- xide sensitive mutant 12 (hpsl2) was screened from this library. Phenotype analysis indicated that hpsl2 plant was dwarf, and the pod was shorter than wild type (WT). The cotyledon greening rate of hpsl2 de- creased after treated with 4 mmol/L H202. Furthermore, the isolated leaves of the hpsl2 mutant showed more obvious symptoms of senescence than WT under treatment of 10 mmol/L H2o2. Genetic analysis suggested that hpsl2 was a single recessive mutant.
