采用超滤—离子层析法精制猪硫酸软骨素,离子对反相高效液相色谱法测定其含量,红外吸收光谱法测定其结构。研究了超滤液温度、超滤压力、超滤液pH对猪硫酸软骨素精制时膜通量和得率的影响。结果表明:超滤—离子层析技术可应用于精制猪硫酸软骨素。猪硫酸软骨素超滤的工艺条件:以微孔滤膜处理原料液,超滤膜的截留分子量10 kD,超滤压力0.25 MPa,超滤温度25℃,溶液pH为9。产物纯度为95.74%,产率为80.15%。该方法精确度和准确度都较高,可用于猪硫酸软骨素的精制。
Porcine chondroitin sulfate was refined by UF membrane-ion-exchange-chromatography.Its structure was determined with infrared spectroscopy and its content was determined by Ion-pair RP—HPLC.The effects of ultrafiltrate temperature,ultrafiltration pressure and ultrafiltrate pH on the flux and yield of porcine chondroitin sulfate were studied when it was refined.UF membrane-ion-exchange chromatography can be used to refine porcine chondroitin sulfate.The process conditions of the refinement of porcine chondroitin sulfate: using microporous membrane to process raw material liquid,the molecular weight cutoff of UF membrane is 10 kD,ultrafiltrate pressure 0.25 MPa,ultrafiltrate temperature 25 ℃ and pH 9.Purity of products is 95.74% and yield 80.15%.This method is of high accuracy and can be used to refine porcine chondroitin sulfate.
