目的:分析NF-κB,Gadd45γ蛋白在肝细胞性肝癌中的差异表达,探讨其在肝癌尤其是HBV感染型肝癌发生发展中的意义。方法:应用免疫组化方法检测NF-κB,Gadd45γ蛋白在64例肝癌和48例癌旁组织中的表达情况,并结合HBV感染状态与临床病理资料进行分析。结果:NF-κB在感染型肝癌组织中的表达率高于癌旁组织和非感染型肝癌。Gadd45γ在感染型肝癌组织中的表达率低于癌旁组织和非感染型肝癌组织,差别具有统计学意义(P〈0.05)。NF-κB的表达与肝癌的分化程度,转移能力有关;NF-κB,Gadd45γ表达呈负相关(P〈0.05)。结论:HBV感染型肝癌组织中NF-κB的高表达,Gadd45γ的低表达并NF-κB部分介导的Gadd45γ表达下调共同参与了肿瘤的发生发展。
Objective:To study the differential expression of NF-κB,Gadd45γ in hepatocellular carcinoma and validate the significance in the tumorigenesis and progression.Methods:The expression of NF-κB,Gadd45γ in 64 hepatocellular carcinoma tissues and 48 adjacent non-neoplastic tissus were detected with immunohistochemical maxvision method,and the results were anaiyzed combined with condiction of HBV infection and clinical pathologic characteristics.Results:In contrast with the expression of Gadd45γ,the positive rate of NF-κB in HBV-infected hepatocellular carcinoma tissues were higher than that in adjacent non-neoplastic tissus and non HBV-infected hepatocellular carcinoma tissues(P〈0.05).The expression of NF-κB was correlated with differentiation of tumor and metastasis.In addition,There was a negative correlation between the expression of NF-κB and Gadd45γ(P〈0.05).Conclusion:The high expression of NF-κB and the low expression of Gadd45γ in hepatocellular carcinoma especially in HBV-infected hepatocellular carcinoma may play a crucial role in the tumorigenesis and progression.
