目的探讨外周抗凝全血中miRNA的表达与维吾尔族肥胖人群的关系。方法按照亚太地区推荐对肥胖诊断的标准,选择维吾尔族肥胖与非肥胖组抗凝全血样本各5例,采用芯片筛选出差异表达的miRNA;后扩大样本量,测量身高、体重、腰围、臀围后,留取血样检测空腹血糖、血脂等指标,采用实时PCR验证差异表达的miRNA。结果芯片结果显示,与非肥胖组相比维吾尔族肥胖组抗凝全血中miRNA130a的表达量下降[(1.14±O.32)vs(0.28±0.13),P〈0.05];维吾尔族肥胖组体重指数、腰围、臀围以及收缩压均大于非肥胖组,并且差异有统计学意义(P〈0.05);实时PCR验证结果显示,与非肥胖组相比维吾尔族肥胖组抗凝全血中miRNAi30a表达量降低[(4.84±1.18)vs(2.03±0.59),P〈0.05];相关分析结果在维吾尔族抗凝全血中miRNA130a的表达量与空腹血糖值、总胆固醇值呈负相关(P〈0.05),而与高密度脂蛋白胆固醇呈正相关(P〈0.05)。结论抗凝全血中miRNA130a的表达量在维吾尔族肥胖人群中显著降低,提示miRNA130a表达量降低可能与维吾尔族BMI有关。
Objective To investigate the expression of microRNA (miRNA)in peripheral anticoagulant blood and its relationship with obesity in Uygur population. Methods 5 obesity and 5 age and sex matched non-obesity from Uygur population were enrolled, differential expression of miRNA of peripheral anticoagulant blood were performed by microarray analysis, then 12 of anticoagulant blood samples with obesity, 16 samples with non-obesity from Uygur population were enrolled. Data on height, body weight, waist circumference (WC) and hip circumference (HC) were collected. Fasting plasma glucose (FPG), triglyceride (TG) ,total cholesterol (TCH), high density lipoprotein cholesterol ( HDL-C), low density lipoprotein cholesterol (LDL-C) were determined. And further miRNA was confirmed by quantitative real- time RT-PCR. Results The expression of miRNA130a in anticoagulant blood with obesity group was decreased compared with that of non-obesity group in Uygur population (0. 28 ~ O, 13 vs 1.14 ~ 0. 32, P 〈 0.05 ) in microarray analysis. The data with body-mass index( BMI), WC, HC and systolic blood pressure (SBP) in Uygur obesity group were higher than those in nonobesity group( P 〈 O. 05 ). The level of miR- NA130a expression in obesity group was lower than that of non-obesity group(2. 03 ± 0. 59 vs 4. 84 ± 1.18, P 〈 0. 05 ) in real-time RT-PCR. Partial correlation analysis showed that the level of miRNA130a expression in anticoagulant blood in Uygnr population was negative correlated with the levels of FPG and TC, and it was positively correlated with the level of HDL-C. Conclusions miRNA130a expression in peripheral anticoagulant blood in Uygur obesity population has significantly reduced, miRNA130a expression might be related to Uygur BMI.
