目的:研究醒脑静注射液对酒精性昏迷大鼠外侧下丘脑区(LHA)谷氨酸和γ-氨基丁酸的影响。方法:腹腔注射乙醇制作酒精性昏迷大鼠模型,20min后侧脑室给药,微透析技术收集透析样品(每30 min收集一次),OPA柱前衍生化法处理样品,高效液相色谱(Synergi Hydro-RP C18,150 mm×4.6 mm,4μm),荧光检测脑脊液中谷氨酸(Glu)、γ-氨基丁酸(GABA)的含量。结果:与模型对照组比较,醒脑静注射液0.34 mg/kg、0.68 mg/kg、1.37 mg/kg外侧下丘脑区(LHA)Glu、GABA恢复到正常水平的时间更短。结论:醒脑静注射液能够通过调节酒精性昏迷大鼠外侧下丘脑区(LHA)Glu、GABA的含量,使其恢复到正常水平从而促进觉醒。
To study the effect of Xingnaojing Injection on glutamate and GABA in lateral hypothalamic area of alcoholic coma rat. Methods: Producing alcoholic coma rat models by intraperitoneal injection and intracerebroventricular administration 20 minutes later, sam- ples were collected by microdialysis (lateral hypothalamic area, collected every 30 minutes). O-phthaldialdehyde was used for the derivatiza-tion of the samples, then HPLC-FLU( Synergl Hydro-RP C18,150 mm × 4.6mm,4 μm)was used to measure the levels of glutamate and GA- BA in dialysis fluid. Results: Comparing with the model control group, the levels of glutamate and GABA in lateral hypothalamic area of ex- perimental group(0.34 mg/kg,0. 68 mg/kg, 1.37 mg/kg)was found to be recover in a shorter time. Conclusions: Xingnaojing Injection could promote arousal of alcoholic coma rats by modulating the content of glutamate and GABA in LHA to normal level.
