目的观察透骨消痛胶囊干预骨性关节炎软骨下骨重塑的作用及机制。方法128只新西兰大白兔随机分4组,除正常组外均按改良Hulth法造模。正常组、模型组予生理盐水10mL/d灌胃,对照组予壮骨关节丸水溶液10mL/d灌胃,实验组予透骨消痛胶囊水溶液10mL/d灌胃,术后分批分次处死取材。RT—PCR法检测软骨下骨c—fos、β-Catenin、RAN甩和软骨MMP-1mRNA的表达。结果与正常组比较,造模后6周、9周、12周,实验组各指标表达均较高(P〈0.05)。与模型组比较,造模后1周.实验组c—fosmKNA表达较低(P〈0.05);造模后6周,实验组RANKL、MMP-1mRNA表达较低(P〈0.05);造模后9周和12周,实验组各项指标表达均较低(P〈0.05)。与对照组比较,造模1周后.实验组c—fosmRNA表达较低(P〈O.05);6周时实验组RANKL、MMP-1mRNA表达较低(P〈0.05);9周、12周后,实验组β—Catenin、MMP-1mRNA表达较低(P〈0.05)。结论透骨消痛胶囊可改变软骨下骨重塑的速率和模式,最终减轻软骨下骨硬化。
Objective To observe the effect of Tougu Xiaotong granula (TGXT) on subchondral bone remodeling and investigate the mechanism. Methods 128 New Zealand rabbits were divided into 4 groups, i. e, normal group, model group, control group and experimental group, 32 rabbits per-group. Except for the normal group, the other three groups were operated on according to the improved method of Hulth. The normal group and the model group were perfused with normal saline 10 ml/d, at the same time the control group and the experimental group were respectively perfused with the water-solution of Zhuanggu Guanjie pill 10 ml/d and TXG 10 ml/d. After operation, each group was executed respectively in 1, 6, 9 and 12 weeks. To observe the expression of c-fos, β-catenin and RANKL mRNA in subchondral bone and MMP-1 mRNA in cartilage which were tested by RT-PCR. Results By the end of the first, the sixth and the ninth week after operation, the expression of each index increased in the experimental group compared with the normal group (P〈0.05). By the end of the first week after operation, the expression of c-fos mRNA was significantly down-regulated compared with the model group (P〈0.05); by the end of the sixth week after operation, the expression of RANKL and MMP-1 mRNA decreased significantly (P〈0.05); by the end of the ninth and twelfth week after operation, the expression of all the molecular biology index was down-regulated in the ex perimental group (P〈0.05). By the end of the first week after operation, the expression of c-fos mRNA in the experimental group was significantly lower compared with the control group (P〈0.05); by the end of the sixth week after operation, the expression of RANKL and MMP-1 mRNA decreased significantly (P〈0.05); by the end of the ninth and the twelfth week, the expression of β-catenin and MMP-1 mRNA in the experi mental group was significantly lower than the other group (P〈0.05).
