番茄褪绿病毒(Tomato chlorosis virus,ToCV)是一种由粉虱传播的RNA病毒,可以侵染番茄和辣椒等常见蔬菜,已在世界多地造成严重危害.2012年在北京市大兴区调查蔬菜病毒病时发现保护地栽培的甜椒植株表现脉间褪绿症状,且植株上烟粉虱数量多,初步推测为ToCV危害.通过提取显症样品总RNA,利用ToCV特异性引物进行反转录PCR,扩增得到463 bp的目标条带.通过测序和核苷酸序列比对分析,表明该序列与国外已报道的ToCV HSP70h(the heat shock protein 70 homolog,热激蛋白70家族)基因序列相似性为99%.这是对ToCV在我国侵染甜椒的首次报道.
Tomato chlorosis virus (ToCV), a RNA virus transmitted by whiteflies, induces severe damage to to- mato and pepper production in the world. In a survey of virus disease on vegetables in 2012, a yellowing disease was found on greenhouse grown pepper plants in Daxing district, Beijing. Meanwhile, a large number of white- flies (Bemisia tabaci )were found on pepper plants. We suspected the diseased pepper plants were infected by ToCV. A Reverse Transcript-PCR assay was performed with total RNA isolated from diseased samples using spe- cific primers for ToCV. Amplified DNA fragments (463 bp) were obtained. The nucleotide sequences of the frag- ments had the highest identity (99%) with that of ToCV. To our knowledge, this is the first report of ToCV on pepper in China.
