通过鸭肝炎病毒F株的测序分析研究了鸭肝炎病毒基因组间的变异情况,为鸭肝炎病毒病的防治提供线索。结果表明DHV-F具有DHV-Ⅰ所具有的基本特征此外还有如下特点:1)DHV-F 5′UTR含有8个茎环结构,且有2个GNRA基序、A/C富含区、U/C富含区、A富含环和U富含环;2)2C蛋白基序DDLxQ中的亮氨酸被苯丙氨酸(192~196 aa)所替代;3)3D蛋白基序PSG中的脯氨酸被半胱氨酸(280~282 aa)所替代;4)预测表明3D蛋白具有“指、拇指和掌”的空间构象,且具有一个大的开放性的中间槽;5)3D蛋白所有保守基序位于蛋白亚基的内层;6)DHV-F的多聚蛋白P2和P3与DHV-Ⅰ的相应蛋白片段有较高的同源性,而与P1的同源性相对较低。说明鸭肝炎病毒DHV-F有抗原性变异。
This study was performed to analyze the status of genome variations among different viral strains for practical clues for the prevention and treatment of duck viral hepatitis. The genome of DHV-F was sequenced and analyzed and found to have typical picornavirus organization and unique properties as reported. In addition, several features were found as follows: 1)the 5'UTR of DHV-F contains eight stem-loops with two GNRA motifs, A/C and U/C rich regions and A-and U-rich Ioops; 2)The leucine in motif, DDLxQ of 2C are replaced by phenylalanine at amino acid 192- 1963 3)The proline in motif, PSG of 3D protein is replaced by cysteine at amino acids 280 - 282; 4)the 3D protein has a conformation with fingers, thumb and palm domains, and a large and exposed central clef; 5)All conserved motifs in 3D protein are located in the inner layer of subdomains; 6)DHV-F has relatively high similarity with other DHV-1 strains in regions P2 and P3, and relatively lower similarity in region of VP1 by SimPlot analysis. These indicate that DHV-F has antigenic variations.
