目的研究CD4^+CD25^+调节性T细胞与慢性特发性血小板减少性紫癜(ITP)的关系。方法用流式细胞分析仪分析来自健康人和ITP患者外周血细胞中的调节性T细胞(Treg)数目及比例;BrDU酶联免疫吸附测定试剂盒测定淋巴细胞增殖。结果活动期和治疗未缓解的ITP患者调节性T细胞的百分比明显低于缓解阶段的ITP患者和对照组;ITP患者体内调节性T细胞对淋巴细胞增殖的抑制活性也存在缺陷。结论ITP患者体内调节性T细胞数量下降和功能改变可能与该病免疫发病机制有关。
Objective To explore the profile and function of CD4^+CD25^+ regulatory T cells (Treg cells) in patients with idiopathic thrombocytopenic purpura (ITP). Methods Treg cell numbers were analyzed by flow cytometric analysis in peripheral blood mononuclear cells collected from control donors and patients with ITP. Quantification of cell proliferation was assayed by an BrdU ELISA kit. Results The percentage of Treg cells was significantly decreased in ITP patients in active and non-remission state when compared to the patients in remission and to healthy subjects. The suppressive activity of Treg cells in ITP patients was also found to be impaired. Conclusion Decreased number and function of Treg cells may be one of mechanisms of immune regulation dysfunction in ITP.
