通过建立新疆陆地棉品种‘新陆早36号’的体外胚珠培养棉纤维体系,利用图像数字化处理技术和电镜切片观察,探索体外培养条件下不同浓度外源阿魏酸(FA)对棉纤维生长发育的影响。结果表明:(1)随着培养时间的延长,培养棉胚珠的体积、棉纤维长度、纤维团生长面积和纤维细胞壁厚度均显著增加。(2)FA的影响在培养15 d后达到显著水平,25和50μmol/L外源FA对离体培养棉纤维的伸长和纤维细胞壁的增厚都具有促进作用,但不同浓度下纤维伸长和细胞壁增厚的程度不同。(3)各处理培养25 d后,25μmol/L FA处理的棉纤维长度比对照极显著增加14.2%,纤维团生长面积比对照极显著增加18%;50μmol/L FA处理的棉纤维细胞壁厚度比对照极显著增加61.7%。
By founded an ovlue culture system in vitro with 'Xinluzao 36' which planted in Xinjiang,we studied the effects of exogenous ferelic acid(FA) in different concentrations on cotton fiber development using the technology of image digitization and electron microscopic observation.The results indicated that all the morphology index including ovular volume,fiber length,growing area and cell wall thickness increased.The effects of FA waxed after 15 day culture.FA concentration at 25 μmol/L and 50 μmol/L can both increase the fiber growth and cell wall thickness,but the incremental extents were seperated.Ovlues were observed and analyzed after 25 day culture,the data revealed that FA concentration at 25 μmol/L can promote the fiber elongation to increase 14.2%(P0.01) and the growth area to increase 18%(P0.01).While conspicuous cell wall thickening was observed when FA concentration raised up to 50 μmol/L,compared with the control,the thickness of the fiber cell wall raised 61.7%.
