本研究利用血清Ⅰ型马立克氏病病毒(Marek's disease virus serotype 1,MDV-1)被膜蛋白UL11原核表达产物作为免疫原,免疫小鼠制备多抗血清,通过间接免疫荧光试验,结果发现:UL11真核表达时,存在于COS-1和Sf9细胞浆中。而检测MDV的RB1B毒株感染CEF时,却发现UL11高度聚集于空斑中心,且可分布于空斑周围的所有细胞核中。这为研究UL11细胞核内扩散的分子机制,以明确UL11是否参与MDV-1致瘤的过程提供了参考。
In the present study,UL11,one of tegument proteins of Marek's disease virus serotype 1(MDV-1),was amplified and expressed in a prokaryotic form and then separated by SDS-PAGE analysis.The target band was collected and used as antigens to immunize mice.In the fluorescence testing using the murine antisera,UL11 was found to be localized within cytoplasma of COS-1 transfected with UL11 and Sf 9 cells infected with recombinant Bac-to-Bac Baculovirus.In chicken embryo fibroblast cells(CEF) infected with strain RB1B,UL11 was accumulated in the center of the plagues and spread into all the nuclei of bystanding cells.Future study will focus on the molecular mechanisms and functions of UL11 and its role in tumor-formation by MDV-1.
