采用水蒸汽蒸馏法提取盾叶薯蓣内生真菌Dzf12的挥发性成分,得率为菌丝鲜重的0.276%。用GC-MS鉴定出30个成分,占挥发性成分总量的95.47%,其中含量较高的有棕榈酸甲酯(22.63%)、(E)-9-十八碳烯酸甲酯(8.30%)、(E,E)-9,12-十八碳二烯酸甲酯(8.12%)、2,6-二叔丁基对甲基苯酚(6.79%)、正十四烷(6.69%)、正十二烷(5.88%)等。测定了该挥发性成分对8种细菌的抑制活性,最低抑制浓度(MIC)值在0.75mg/mL至2.00mg/mL之间,抑制中浓度(IC50)值在0.62mg/mL和1.55mg/mL之间。对番茄疮痂病菌的抑制活性最强,MIC和IC50值分别为0.75mg/mL和0.62mg/mL。
The volatile oil was extracted with a yield of 0.276% on fresh weight basis by hydro-distillation from Dzf12,an endophytic fungus isolated from Dioscorea zingiberensis.The oil was analyzed for its chemical compositions by gas chromatography-mass spectrometry(GC-MS).Methyl hexadecanoate(22.63%),methyl(E)-9-octadecenoate(8.30%),methyl(E,E)-9,12-octadecadienoate(8.12%),2,6-di-tert-butyl-4-methylphenol(6.79%),tetradecane(6.69%),and dodecane(5.88%)were the major compounds of the 30 identified components which accounted for 95.47% of the total volatile oil.The in vitro antibacterial activities of the oil on eight bacteria were investigated.The minimum inhibitory concentration(MIC)and median inhibitory concentration(IC50)values of the oil against the test bacteria ranged from 0.75 mg/mL to 2.00 mg/mL and from 0.62 mg/mL to 1.55 mg/mL,respectively.One of the most sensitive bacteria was Xanthomonas vesicatoria with the MIC and IC50 values to be 0.75 mg/mL and 0.62 mg/mL,respectively.These results indicated that the endophytic fungus Dzf12 would be a candidate for producing biodiesel as well as a potential source for producing antimicrobial components.
