目的应用PCR-DGGE(denaturing gradient gel electrophoresis,变性梯度凝胶电泳)技术对孤独症家庭成员肠道微生物菌群结构进行研究.材料和方法提取粪便细菌总基因组DNA,PCR扩增细菌16S rDNA基因V3可变区,DGGE方法检测PCR产物.结果建立了孤独症家庭成员肠道菌群组成的DGGE指纹图谱,分析结果显示孤独症儿童肠道菌群结构的相似度为0.42-0.65,家庭内孤独症儿童与父母之间的相似度为0.42-0.68.结论 PCR-DGGE技术是一种快速有效的用于分析研究人体肠道菌群结构的技术.孤独症儿童肠道菌群结构与父母的相似度较高,提示生活环境对肠道菌群的组成有影响.
PCR-DGGE(denaturing gradient gel electrophoresis) was used to analyze the diversities of intestinal bacteria in sixteen individuals from seven autism families.DGGE analysis showed that the similarity values of the comparisons of the bacterial DGGE bands patterns of the autistic individuals ranged from 0.42 to 0.65,and the autistic children and their parents in the family ranged from 0.42 to 0.68.PCR-DGGE was proved to be a powerful tool to analyze the intestinal bacterial diversity.There existed a correlation between the intestinal bacteria and the living environment.
