Swine are more analogous to humans than rodents in anatomy and physiology. Comparing with non-human primates, pigs are more easily available and have less ethical concern. Therefore pigs have more extensive applications in the human health related research field. In this review, we mainly summarize the progress of generation of genetically-modified pigs in modeling various human diseases and xenotransplantation. We also describe some achievements on a project about generation of genetically-modified pigs for reproduction and development funded by Ministry of Science and Technology of China.
