采用ISSR分子标记检测海南岛青梅(Vaticamangachapoi)7个自然居群192株个体的遗传多样性及遗传结构。筛选的10条ISSR引物共扩增出清晰位点132个,其中多态性位点130个,多态性百分率(PPB)达98.48%。物种水平上,Nei’s基因多样性指数(H)为0.3391,Shannon多态性信息指数(I)为0.5095,总基因多样度(Ht)为0.3441,反映了较高的遗传多样性。AMOVA分析显示,青梅的遗传变异主要存在于居群内个体间,但亦产生了相当程度的居群间遗传分化,Nei’s基因分化系数Gst为0.3329。Mantel检测表明,青梅的遗传距离与地理距离间没有显著相关性(r2〈0.2),地理隔离对青梅各自然居群间的基因流影响不明显。
Genetic variation and structure of 192 individuals of Vatica mangachapoi from seven natural populations in Hainan Island were investigated by using inter-simple sequence repeats ( ISSR ) method. Amplification with ten ISSR primers generated 132 unambiguous loci, 130 of which were polymorphic indicating 98.48% of polymorphic loci. High levels of genetic diversities were detected in V. mangachapoi at the species level with 0.339 1 Nei's genetic diversity ( 0.509 5 Shannon information index and 0.344 1 total genetic diversity ) . The Nei's coefficient differentiation ( Gst ) by ISSR was 0.332 9, suggesting that the main genetic variations of V. mangachapoi were located within populations and moderate level of differentiation among populations. The Mantel test analysis showed that there was no significant correlation between genetic distance and geographic distance ( r2〈0.2 ) , implying limited influence of geographic isolation on the gene flow among populations.
