[目的]研究束缚-低氧交叉适应大鼠心功能变化及与心肌HSP70表达水平间的关系。[方法]雄性Wistar大鼠随机分为对照组、低氧应激组、束缚适应组和低氧适应组。通过检测血浆肾上腺素浓度,评价交叉适应模型的建立;测定各组经低氧暴露后的心率、心电图、血清LDH活性,从而评价大鼠心功能的变化。同时测定各组中心肌细胞HSP70表达水平,并分析HSP70与大鼠血清LDH活性的相关性。[结果]交叉适应组大鼠的心率、异常心电图率和血清LDH活性均明显低于低氧应激组,而心肌HSP70表达水平明显高于低氧应激组。HSP70表达水平与大鼠血清LDH活性具有负相关性。[结论]束缚适应可通过提高机体的心功能产生对低氧应激的保护作用,交叉适应的建立与HSP70在细胞中的表达量相关。
[Objective]To investigate the association between the cardiac function change of restraint-hypoxia cross-adaptation and HSP70 expression in rats.[Methods]Wistar rats were divided into four levels randomly:control group,hypoxia stress group,restraint adaptation group and hypoxia adaptation group.The concentration of adrenaline on plasma was detected to evaluate the establishment of restraint-hypoxia cross-adapted rat model.After acute hypoxia exposure,heart rates,ECG and LDH activity in blood serum of each group were measured for evaluation of cardiac function.Equally,HSP70 expression of cardiac muscle cell were detected in each group,and analyze its correlation with the LDH activity in blood serum of rats.[Results]When compared with the hypoxia group,ECG and LDH activity in adaptation group were significantly decreased,while HSP70 expression was significantly increased.The HSP70 expression showed inverse correlation with LDH activity in blood serum of rats.[Conclusion]Restraint adaptation can improve the protection towards acute hypoxia by enhancing heart functions,and the establishment of cross adaptation is correlated with the levels of HSP70 expression in cardiac muscle cell.
