目的研究纳米化蒙脱石衍生物对甲型流感病毒的吸附作用,了解其能否阻止病毒侵入细胞。方法将梯度稀释的纳米化蒙脱石衍生物PTK—RY00101001分别与PR8、Memphis、Aichi3种病毒结合后感染MDCK细胞,利用细胞感染免疫染色方法测定PTK-RY00101001对病毒感染细胞的作用,并通过测定MTr观察PTK-RY00101001对正常细胞的损伤影响。此外,将不同浓度PTK-RY00101001固定的纱布与流感病毒作用.对纱布和剩余病毒液分别进行免疫实验和细胞感染免疫实验,判定PTK—RY00101001对病毒的吸附作用。结果PTK-RY0010100l在25μg/mL以下,对正常细胞几乎无损伤。细胞感染免疫染色试验显示,PTK-RY00101001对流感病毒感染细胞的能力有较强减弱作用。不同浓度PTK-RY00101001固定的纱布与病毒作用后,纱布染色结果显示无明显差异,通过剩余病毒液感染细胞免疫实验,1000和200μg/mL浓度的PTK-RY00101001与纱布固定后,PTK-RY00101001对病毒有一定的吸附作用。结论纳米化蒙脱石衍生物在不损伤正常细胞的浓度下,具有通过吸附甲型流感病毒,从而阻断病毒感染细胞的能力,因此推断其能通过吸附作用而阻止流感病毒入侵细胞。
Objective To study the adsorption of influenza A virus by nano-montmorillonite, and judge whether it can block the influenza virus from invading ceils or not. Methods The different concentration of nano- montmorillonite (PTK-RY00101001) were combined with 3 kinds of viruses (PR8, Memphis, Aichi strain), then infected MDCK. The effect of PTK-RY00101001 for influenza virus infecting MDCK cells were evaluated by cell immunostaining experiments,and the damage effect of PTK-RY00101001 on normal cells was detected by MTT method. In addition, the gauze which was fixed with different concentration of PTK-RY00101001 interacted with influenza virus, then the guaze and remainder virus were studied by means of immunization experiments and cell immunostaining experiments to determine whether the PTK-RY00101001 could prevent the virus invasion or not. Results Under the concentration of 25 μg/mL, PTK-RY00101001 could hardly damage normal cells. The cell immunostaining experiment result showed PTK-RY00101001 had a weakening effect on the ability of influenza virus to infect cells. After the gauze which was fixed with different concentration of PTK-RY00101001 interacted with influenza virus, the results showed no significant difference. However, after the remaining virus-infected cells immunization experiments, the PTK-RY00101001 concentration in 1 000 and 200 μg/mL had a certain adsorption on virus. Conclusions In the concentration of montmorillonite that does not damage normal cells, it can block the ability of the virus from infecting cells by adsorption of influenza virus. Therefore, it can be inferred that montmorillonite can prevent the influenza virus from infecting cells by adsorption.
